1. Please fill out the internal sample submission.doc or external sample submission.doc prior to submitting samples so that the facility may obtain all the proper materials for processing the samples. Forms can be emailed to lcraddoc@wfubmc.edu or dropped off at the facility in Gray Building Room 3089.
2. Contact Lou Craddock by email or phone (713-5103) to discuss the time frame of completing the work.
3. Please provide one of the following in a 1.5 ml RNase-free microcentrifuge tube:
a) At least 15µg of high quality total RNA in 50-100µl DEPC-treated water. Total RNA should be prepared using TRIzol. Samples should have a A260/A280 ratio between 1.8 and 2.1, and have clearly distinguished 18S and 28S bands on an agarose gel.
b) At least 30µg of biotin labeled cRNA in DEPC-treated water. The facility cannot assume responsibility for the results of the experiment
c) Small samples of 1-2µg of total RNA in DEPC-treated water. Due to the small amount of sample, the facility will not be able to run proper quality control checks and cannot assume responsibility for poor quality arrays.
d) Fully prepared hybridization cocktail prepared from Affymetrix Eukaryotic Hybridization Target Preparation. These samples will not be tested for quality; the facility cannot assume responsibility for poor resulting arrays.
4. All samples must be clearly labeled with the name, concentration, type of RNA, and PI initials. A note on naming: Name your samples as you would like them to appear on the data files, keeping in mind that the name must follow Windows2000 file naming conventions. Also, note which sample is the control sample (baseline) if you are requesting any comparison analysis.
5. Quality control checks will be run on all samples (with the exception noted in 3c and 3d). We will proceed with the process only if the quality and quantity of RNA is acceptable.
