Analytical Imaging Core Laboratory

Laboratory purpose:
A multi-purpose imaging facility for acquisition and analysis of images generated from 2-D radionuclide tracer samples (Phosphorimager), fluorescent tracer samples (Typhoon Imager) or images acquired by scanner or CCD video camera input (Alpha Innotech gel documentation station).

Laboratory capabilities:

  • Imaging for quantitative and qualitative analysis of 2-D radioactive or fluorescent samples such as blots or gels, labeled with 32P, 35S, 14C, 125I, 3H, or with tags for fluorescent or chemiluminescent detection, with 3-5 logs of dynamic range.
  • Scanner or video camera-based image acquisition, storage, analysis, and printing for documentation of SDS-PAGE and ethidium bromide-stained DNA and RNA gels.
  • Multiparametric flow cytometry and cell sorting.
  • Generation of cDNA macroarray blots for expression analysis.

Laboratory services:

A part-time technician is available by appointment to train users in the use of self-serve imaging equipment and software (ext. 6-6347, leave message). Temporary data storage (up to 4 months) is allowed on PC hard drives, with at least monthly backup to tape. Users are notified prior to data purging to free hard drive space.

Instrumentation:

  • Molecular Dynamics Phosphorimager 445SI (radioactivity detection only)
  • MD Typhoon 8600 scanner (radioactive, fluorescent, or chemiluminescent labels)
  • Alpha Innotech IS-2000 imaging workstation for gel documentation and analysis
  • UMAX Astra 4000 flatbed scanner (1200 dpi) for 2-D color or grayscale imaging
  • 35mm slidemaker for direct color image capture to film (e.g. from PowerPoint)
  • Two Dell pentium/windows NT workstations with image manipulation and analysis software and a Macintosh iMac with image analysis software and zip drive
  • Becton-Dickinson FACStar Plus flow cytometer/cell sorter
  • Genomics Solutions FlexSys robotic array spotter
